Once it's "lost" just go on to the other towns that need saving. The "lost" town will eventually have people in it again (assuming you continue the story/questline), which you can save. Besides, saving towns doesn't really affect the outcome of the game. It's all about completing quests, getting trophies, etc.
Yes you can play them on most game types but they will not be able to play undead overrun with you unless they have the undead nightmare game/dlc as well.
