You must go through the temple when it is right side up first, then you will receive the light arrow in the end. Next, go outside the temple and move the center block out of the way and freeze it with your statue song. Look at the place where the block was and you will see a giant red ruby. Shoot this with the light arrow to turn upside down.
Unlike other Zelda games, it is possible to do a vertical spin to flip a skulltula. A vertical spin is performed the spin way as the horizontal one; just swing the wii remote and the nunchuks vertically rather than horizontally. If you swing the nunchuk and wii remote upwards, Link will do a backflip and swing his sword at the same time, performing a horizontal spin that can flip a skulltula. If you swing the wii remote and nunchuck downwards, Link will do a frontflip and swing his sword at the same time, thus, once again performing a vertical spin that CANNOT flip the skulltula. That is the easiest method of flipping a skulltula although you can also do a simple vertical strike with your sword. Just swing the wii remote upwards twice in a row and the skulltula will be flipped.
you have to use a skyward strike but instead of doing it over the skulltula u have to flip it over onto its back but slicing it under the skulltula then use the ending blow on its glowing spot to defeat it.
Put a bomb down near it until the bomb explodes, this should flip the spider over which will let you do a Fatal Blow on it and kill it. Also if you don't have bombs wait for it to try jumping on you, immediately do a Shield Bash (shack the Nunchuk) to knock it on it's back and do a Fatal Blow to kill it.
If they are hanging, swig your sword sideways to spin them around and then stab the purple spot on their belly. If they are chasing you on the ground swing upwards to flip them on their back then use a fatal strike (jumping attack) to stab the same weak spot. If you don't know how to do Fatal Strike then listen. When your fighting, if you knock someone down or flip someone over you would hold z and you would see a diagram of the wii remote and nunchuck. While continueing to hold z swing both the nunchuck and wii remote up.

You need to use the hammer. It depends which part of the temple on the isle of ruins you are talking about. You don't have to hit on the tiles themselves, you can hit the edges and it will flip the tile next to it. Whichever tile you hit, the tiles surrounding it will flip.
