The Missouri Compromise did keep the nation together, but after the Mexican war, the nation got bigger!

A new compromise had to be worked out, and this one didn't succeed.Part of it was theFugitive Slave Act, which aroused Abolitionist sympathies in the North, so the prospect of new slave-states was more remote than ever, and the South had more reason to secede.
