No. Puerto Rico is the smallest island of the Greater Antilles. It could be argued that the Puerto Rican islands of Culebra and Vieques begin the chain of the Lesser Antilles, but it is generally accepted that St. Thomas (east of Puerto Rico and its eastern islands) is the closest of the Lesser Antilles islands to Puerto Rico and its outlying islands.
