As Alexander was a world conquer and was never defeated in battle. He spread Hellenstic culture, the Greek culture.
The most important centre of commerce and Hellenistic culture and the largest city was Alexandria of Egypt. There were also other great centres. The other major ones were Antioch (Syria) Pergamon, Ephesus and Smyrna (Turkey) and Athens.
There was a superficial blend of Macedonian, Greek and Egyptian culture amongst the ruling Macedonian-Greek aristocracy. The Egyptians continued with their own culture.
It was the Center of helenistic Culture as well as the center of commerce during that time period

Its upper class of Macedonians and Greeks which took over the city during Alexander the Great's invasion established Greek customs and culture. Part of this was art and literature promoted and financed by the Macedonian Ptolemies who had made themselves pharaohs. So Alexandria became a leading centre with the largest library and associated scholars.
Alexandria is in Lower Egypt, because it is in the Nile Delta. However, Alexandria only comes into existence late in Egyptian history, long after the most famous Pharaohs and long after there was a significant distinction between Upper and Lower Egypt in terms of culture.
