To find out the qualitative response to changes in Vgs and Vds, consider Vgs =0. In response to a small applied voltage Vds, a n-channel JFET acts as a simple semiconductor resistor and Id increases linearly with Vds. With increasing current, the ohmic voltage drop between the source and the channel region reverse biases the junction and the conducting portion of the channel begins to constrict. because of the ohmic drop along the length of the channel itself, the constriction is not uniform but is more pronounced at distances father from the source. Eventually, a Vds is reached at which the channel is "pinched off". At this voltage, Id begins to level off. and approach a constant value. It is not possible for the channel to close completely and reduce Id to zero. If that were the case, then the ohmic drop required to provide the back biasing would be lacking.
Id is not equal to zero at Pinch off voltage.
Thus, pinch off voltage can be defined as the gate reverse voltage that removes all the free charge from the channel.

I believe you're referring to the triode mode of a MOSFET. In this case, the gate to source voltage, VGS, is greater than the threshold voltage, Vth. The first step is to do a Taylor expansion of the drain current formula:

It will make more sense if you first study the construction of MOSFET. It has three terminals, Gate (G), Source (S), and Drain (D). G is insulated from the S and D. Between S and D is a variable resistor, whose resistance depends on the voltage applied across G and S. There are two primary types of transistors classified on the basis of power: bipolar junction transistors (BJT) and field effect transistors (FET).
Bipolar junction transistors are current-controlled devices, meaning that the amount of current flowing through the device is controlled by the voltage applied to the base electrode. The base-emitter voltage (VBE) and base-collector voltage (VBC) determine the amount of current flowing through the device. The current flowing through the device is given by the following equation:

Where β is the current gain of the transistor.
Field effect transistors are voltage-controlled devices, meaning that the voltage applied to the gate electrode controls the amount of current flowing through the device. Transistor is an active device. Active devices (unlike the passive devices such as resistors and capacitors) you have to bias them so that they can function properly. A resistor behaves well no matter what voltage or current you apply to them (it still obeys I=V/R). But a transistor has to be biased at the right voltages in order to operate as what it is supposed to be.
For example, in bipolar npn, to have it as signal amplifier (e.g. common emitter), VBE (voltage across Base and Emitter) has to be larger than certain voltage (e.g. 0.7V) and VCE (voltage across collector and emitter) has to be larger than another voltage (e.g. 0.2V). Otherwise, the transistor won't be able to amplify the signals with high gain.
In a MOSFET (e.g. NMOS), similarly, we have to bias VGS (gate to source) to be larger than the threshold voltage and VDS (drain to source) to be larger than the saturation voltage.
In conclusion, biasing is to use voltage to set the active element at the "right operating region"! The above are just examples. And depends on what you want the transistors to do, you need different "biasing condition". E.g. if you want to make the MOSFET behave like a resistor, you make VDS smaller than the saturation voltage.
So, after biasing, how can they be used? Usually, a small voltage (signal) will be coupled to the biased transistors for signal processing.

gm = d(ID)/d(VGS), where ID = drain current, and VGS = gate bias, with all other biases fixed and all biases referenced to the source voltage. gm is the measure of how much the drain current changes with an incremental change of VGS. Practically, you measure (ID, VGS) pairs at a fixed VDS for a range of VGS. Then gm at a certain (VGS=VGS1) is simply (ID2-ID1)/(VGS2-VGS1), where 1,2 signify consecutive pairs of (ID, VGS). In other words, you can plot ID versus VGS on an x-y plot with VGS being the x-axis. Whichever way you measure the slope at a certain (VGS=VGS1) on the curve is the gm value.
