Most of the Guitars you will find in music shops and in online music stores are made by large manufacturers. Dean, Epiphone, Fender, Gibson, Gretsch, Guild, Ibanez, Martin, PRS, Rickenbacker, Schecter, Squire, and Taylor are all examples of large guitar manufacturers that you will find just about anywhere. There are also a lot of smaller manufacturers that build guitars.
In these cases, much of the "building" process is handled by machines. There are always a few steps that need to be completed by a person, like the final finish polish on the body or the final set-up of the guitar.
Some guitars are hand made, or mostly hand made. Professional guitar builders are called Luthiers. Some of thesesmaller guitar builders are one-person shops, while others may employ a small number of Luthiers.

Hand made guitars are made totally from scratch. The Luthier chooses the woods to be used and cuts, carves and shapes them into pieces that are needed to build a guitar. Some parts, such as guitar tuning machines or pickups, may be purchased from an outside supplier and added late in the process.

With a hand made guitar, you can specify any aspect of it to your liking. However, it is always worthwhile to seek the advice of the Luthier as he or she has a better sense of what works best.
On a hobby level, anyone can build a guitar if they are so inclined. There are suppliers who will provide complete kits and all the tools you need to do it. If you try it, you may be surprised at the complexities involved in building a quality instrument.
