Get a sturdy container that's appropriate for what you need. Hiking for example it may need to be waterproof; in a car it will need to be sturdy to survive falling over. You can buy bags made just for first aid kits, but anything can do the job.
Put all the supplies you need in it. Keep them organised so they are easy to find. Ziploc bags are an easy way to do this. Include the basics, plus any specialized items you need for your purpose. Hikers will want moleskin for example; asthmatics will want a spare inhaler, etc.
Here are the basics:
Band aids and bandages.
Tape for the bandages.
Antibiotic ointment.
A small roll of stretch gauze.
Alcohol wipes (for disinfecting) (replace yearly, they dry out)
A small medicine container with ibuprofen (for reducing swelling or fevers) and Benadryl (for insect bites and Allergies). Label clearly. Replace yearly.
Tweezers
A small knife and/or scissors. The keychain size Swiss Army Knife works very well.
A felt marker (for writing down times on the bandages or patients skin)
2-4 pairs of latex or nitrile gloves
if you don't know it, don't do it , you will hurt rather than helping.
