In the time of the British American colonies, slavery was an institution on a world wide basis. Colonists in the 13 British colonies shared the views of most of the European world at that time. The view was that Black Africans were inferior beings and that White people were superior to them. In their terms there was nothing wrong with slavery.

The people in the settlement most likely treated the Native Americans badly, and sold some into slavery, as most European colonists did. Therefore, the Native Americans would attack some parts of the settlement, and would refuse to trade with the colonists.
It is beleived that some of the Roanoke colonists may have been taken in by native tribes, however there is no evidence to believe this was a "rescue", the native people were known to take captives and some of the Roanoke colonists may have been kept alive as slaves.

The issue in the civil war is Slavery. I don't believe racism (as in Hitler's Arian race supremacy) is the issue. Although there were some religious under(over?)tone (God approves slavery), Abraham Lincoln, being an Atheist chose to follow the dictates of his conscience and acted to stop the practice of slavery.
