The Prime Minister of the UK preceding Winston Churchill was Neville Chamberlain (Conservative), who came from a very unfortunate political family. Neville's relative Austin Chamberlain was the first Conservative leader to not be Prime Minister and Neville himself became the first Prime Minister to not fight a general election as he took over from Stanley Baldwin in 1936.
Either the cabinet minister has to resign (if the cabinet still supports the Prime Minister), or if the cabinet no longer supports the Prime Minister the Prime Minister has to resign (resulting in a party election) or the Prime Minister can call a general election.
