Ribosomes are the factories of protein synthesis. which remain associated with rough ER. Rough endoplasmic reticulum, is called so due to the appearance of this organelle in electron micrographs. The roughness results from ribosomes, which are studded on the membranes of the organelle. Rough ER, is a network of interconnected flattened sacs, with two main functions. One is to make more membranes so as to accommodate the proteins made by ER enzymes. As a result, the ER membrane enlarges. The other major function of rough ER is to make proteins that are secreted by the cell. An example of such a secretory protein is an antibody, a defensive molecule made and secreted by white blood cells. Ribosomes of the rough ER synthesize the antibody's polypeptides, which assemble into functional proteins inside the ER. The synthesis and packaging of a secretory protein is done by ER and can be understood as following :
1. As the polypeptide is synthesized, it passes into the ER.
2. Short chains of sugar molecule are then linked to the polypeptide, making the molecule a glycoprotein.
3. The ER packages these glycoproteins into tiny sacs called a transport vesicles which buds off from the ER membrane and travels to the Golgi apparatus for further processing. From there, a transport vesicle carry these molecules to the plasma membrane and release its contents from the cell.
