The slope of a line is determined using the coordinates of at least 2 points on the line. If you have 2 points (A, B) and (C, D), the slope of the line can be determined using the formula

(B - D) / (A - C) or (D - B) / (C - A)

which is essentially the difference of the y-coordinates divided by the difference of the x - coordinates.

Alternately, you could think of it as "rise over run", the increase in height (change in y) between the two points over the horizontal distance traveled (change in x).
