Sailor Neptune (Michiru Kaiou/Michelle) and Sailor Uranus (Haruka Ten'ou/Amara) never explicitly reveal that they are in a lesbian relationship together, but it is strongly implied in both the anime and the manga.
By the fourth season/arc of both the anime and the manga, their living together and, in the manga, caring of Hotaru together is not looked on as unusual in any way by any of the other Sailor Senshi or their friends.
In the English dub of the anime, however, Michelle and Amara are said to be cousins, rather than lovers.
Sailor Uranus and Neptune never formally join the Sailor Scouts; throughout the third arc of the manga and the third season of the manga, though Uranus and Neptune occasionally assist the other Sailor Scouts, at times they are actually opposed to one another, as Uranus and Neptune aim to stop the Silence even if it means taking the Talismans from the Pure Heart Crystals of innocent people.
They disappear at the end of the Death Busters arc, only reappearing toward the end (in the manga), and in a special (in the anime). They reappear "for good" as part of the Sailor Team in the fifth and final season of the anime, Sailor Moon Sailor Stars, which concludes the Nehellenia arc from Sailor Moon SuperS, in episode 167, and remain part of the team until their deaths/defection to Shadow Galactica, and rejoin once they are reborn after Sailor Moon defeats Sailor Galaxia/Chaos.

In the original publication run of Sailor Moon, act 25 in volume 8 featured the first appearance of Sailors Uranus and Neptune, along with Chibiusa's meeting of Hotaru. There is no anime episode exactly equivalent to the events shown in this arc, but episodes 90 (Sailor Uranus and Neptune's first, shadowy appearance in the anime) 112 (Hotaru's first appearance and her meeting with Chibiusa) come very close.

The Outer Senshi (Neptune, Uranus, Saturn, and Pluto) do not come in until later in the series, by which time everyone knows that Usagi is the Moon Princess. But the Inner Senshi did not find out until half-way through the first Season (of the anime, I haven't read the manga) that she was the Princess.
Hotaru Tomoe shows up in episode 112, in the Sailor Moon S season. In the subtitled version of Sailor Moon the episode is called "Who is the True Messiah? Chaos of Light and Darkness," and in the dub of Sailor Moon, the episode is called "Show Stoppers."

yes they do die a couple of times. In the sailor stars episode i believe they all die even darien but come back. Darien comes back at the last sailor stars episode for the last five minutes
