Yes, you should have no problem with cake batter that is stored overnight in the fridge. Remove the cake from the fridge at least an hour (longer for a larger cake) so it returns to room temperature before baking.

If you're defrosting it, I would say more than an hour. If you just baked a cake and its hot, I would wait about thirty minutes to an hour before eating it. I'd also say putting it in the refrigerator for 5mins.
