Absolutely! I usually put enough butter into the pan to cover the bottom half of the asparagus. Mix it up a few times while baking and then the asparagus takes on the buttery taste and you have a plate of delicious veggies! The big draw back is that you've now made a healthy dish into something slightly less healthy and slightly more cholesteroly.
It depends on what you're trying to accomplish. If you're going for flavor, I would use butter, but olive oil might be slightly healthier. However, if you use duck fat instead of either, it does basically the same thing as butter, and has about half the cholesterol of butter.

That probably depends on what the frozen asparagus would have had done to it.... if it was to be baked then probably not. Asparagus doesn't require much cooking, and it is likely that if the frozen asparagus was to undergo heating for any length of time then that might be enough time for the fresh asparagus to cook. You might blanche the asparagus first - plunge it into boiling water for a minute before using it in the recipe.
