Curley attacks Lennie because he assumes Lennie is laughing at him when really he is laughing and smiling about the ranch him and George dreamed of. Also because Curley goes after big guys and he thought Lennie wouldn't fight back so that's why he punches him and makes his nose bleed but then George tells Lennie to fight back and Lennie ends up crushing Curley's hand.

Curley attacks Lennie out of misplaced aggression and insecurity. He feels threatened by Lennie's size and strength, and wants to assert dominance over him. Curley's aggressive behavior is also a reflection of his own insecurities and need to prove himself as a tough guy on the ranch.

Curley has a Napoleon Complex, which means he is a little man who feels inadequate and must prove himself tougher than big men. Lennie is a monstrous man, so Curley sees him as a challenge and a conquest.

You can interpret it however you like, I believe that because he was seemingly physically inferior (crazy short) he felt he had to prove to everyone that he was not, by fighting the biggest guy he could find.
