The dealer has to do it with a scan tool.
If it's the original transmitter and it has gone out of sync with the vehicle, hold down both the unlock and lock buttons until the car horn honks. This should re-sync. If it's a replacement transmitter, I think the dealer will have to program it.

After a delay of approximately 15 seconds , the doors will lock and unlock to confirm the programming of that transmitter. # Repeat the previous step to program up to four transmitters. # Turn the ignition switch to the RUN position in order to exit the keyless entry transmitter programming mode. # Operate the transmitter functions in order to verify correct system operation. Method 2 - Scan Tool Required, Program Individual Transmitter Only use this option if you are certain which transmitter #1- #4 is being replaced, or the new transmitter can be programmed into a previously unused position within the PDM memory. If you just want to replace the transmitter battery on it it does not need to be program it does need to be resequenced. 1 replace the transmitter battery 2 close all doors on the car 3 stand outside of car , on the driver side , and hold at the same time the LOCK and UNLOCK buttons, you should hear the locks cycling close to open position. After that your transmitter is resynched, if it does not work you may need a new transmitter. If you do it does need to be programmed at the dealership and you will need the car there along with any other transmitters you have for the car.

I know this works for the X type but you might as well try it and see if it works for you! Here are the instructions: Hold headlight flasher back. Turn ignition key to position 1. Flash headlights 4 times. Alarm will enter programming mode. This will be confirmed by "beeps" from the alarm and flashing led on gearshift. Program each transmitter (up to 5) by pressing the lock or unlock button on each transmitter once. The alarm will "honk" when you press the transmitter button. The alarm must honk 5 times to complete the programming. Therefore, if you are only programming one transmitter you should operate the transmitter 5 times. If you are programming two transmitters you will operate one transmitter twice, and three times on the other.

You can't program the keyless entry remote for the Dodge 2500 yourself. You have to have it done at a Dodge Dealership. The cost is around $75.
