You computer needs an internet connection to update(in most cases), in order for your computer to recive an internet signal, it needs to be turned on. I think it may work if you have a screen saver on, but i am not 100% sure on. A way to test it is to start an update and then put your computer into screen saver mode and wait a couple minutes, if the percent goes up it works :P. Happy Updating
Code 646 basicly means that your firewall is denying access to your computer's FTP ports. Windows update needs FTP ports to download updates. In order to update your computer you may need to temporarily disable any firewall you have running so Microsoft can access your computer. If this doesn't work, you may need to give India a call...

If you download Microsoft Security Essentials and make it scan your computer, it will find viruses that cause the error, and will allow you to remove them. If Microsoft Security Essentials can't run (because it needs to update), look for the update online.
