Download Anti-Virus and install it on your system. Be sure it's updated always. If you're using Windows XP be sure to enable the firewall for additional protection. Actually there is no 100% protection from Trojan since there is an open port when accessing to the internet which will make it vulnerable to trojans.

Depends what type of Trojan it is, if its just on your hard drive, in your documents or something, just delete the folder its in and empty the recycle bin. If its got into your WINDOWS folder (Which the most powerful ones do), then first try a better anti-virus to delete it, i recommend NOD32, if it fails, use unlocker to delete it, if that fails, then you will need a re install of windows.
I used Spy Sweeper, and cleaned one out about 5 minutes ago, but it keeps restarting itself, any suggestions?
to get rid of a Trojan virus u must reboot your whole computer. You cant just get rid of it completely. So just restore it to its original settings:)
As for antivirus software,I suggest you Kingsoft Free Antivirus which ranked second in the recent av-c test. It's really perfect.I've tested it on my own computer.
