I think you've got this wrong do you mean why is Akito so protective over Yuki? If that's what you mean it is because traditionally the rat was the favorite because he arrived at the feast first. Akito is very protective over Yuki because he could as Yuki's parents basically gave Yuki to Akito to gain his favour and so Akito pretty much owned Yuki while he was younger and used this power to make his life misserable.

Based on the memories we've seen in the manga from Yuki's childhood, Akito emotionally abused Yuki. He would tell Yuki that he was worthless, no one appreciated him, and the other zodiac members blame him for their lives being miserable just because he exists. He also locked Yuki in a dark room and whispered these things to him, so he basically tried to brainwash him into thinking these things. You can also see Akito whipping Yuki, while telling him this.

In the anime Akito is introduced as male and remains male. I don`t think Ren, Akito`s mother, even appears in the anime.
