use something like a cuticle stick, or something you can get underneath your nail without damaging it, wrap it in cotton wool and dip this into some nail varnish remover, alternatively you could use a cotton bud dipped in remover. then slip this under the nail and wriggle it till the polish is removed!
I would use polishing compound or even a wax with a cleaner in it. If the car was waxed even a while back, there will still be a coating or barrier between the nail polish and the car finish. A little rubbing and it should come up.

A Sharpie and nail polish remover have the same chemicals in them. Nail polish remover has the chemicals to remove that so it adapts also to Sharpies.
