I regularly drive Sydney to Wilsons Prom to visit family. I have taken just about every route and have found the fastest to be Sydney to Cooma to Cann River to Wilsons Prom. It takes me 11.5 hours with ok traffic (including rest/meal breaks) and I often do it in a single day. However, if you are not used to driving, or not used to country roads or not willing to drive the speed limit, then I'd recommend you have an overnight stop.
