Check your battery first.. If it is fine, you might need to replace your cold start injector timing switch. Fairly expensive, but should work.
Obviously something is frozen. When you say not start do you mean everything is like normal and the engine just refuses to run? OR when you turn the key to start, nothing happens. Usually the normal cold weather villains are #1 BATTERY #2 Contaminated fuel system. #3 Driver Error
User manual for 2000 4Runner suggests 5W40 for extreme cold temperatures for the 3.4 liter V-6. I use 10W40 however live in the SW (Arizona) so temperatures never get cold. Hope this helps?

It stands for the Air Intake Sensor. Basically it is another sensor used by the computer to adjust fuel mixture for a good running car in cold weather. It is housed in the air filter flow assy unit.

Disconnect the battery. It's probally doing it because of the weather change. We had a 1985 Toyota 4Runner back in the day and it'd beep non stop when it'd get cold at night.
There is no power steering in a Mercury Villager at start up in cold weather due to a failing pump or low fluid. Fluid becomes much thicker in cold weather and is much harder to pump.

Your model should have a motor plug. If you plug it in to the wall outlet in cold weather overnight, you should have no problem to get it to start.
