The axle will have a metal nut on the end of it. Usually very rusted and deep inside the hub. Clean out all the old grease and gunk and you should find it. Try penetrating oil to loosen it but usually it takes heat to loosen which burns up the wooden hub badly so do as last resort.
This is not easy, but can be done by a good wood worker. First the iron rim has to be removed by heating it to expand enough to loosen off the wheel. If the feloe is still intact that helps, but it has to be dismantled carefully. The new spoke ( made to match) is then installed first in the hub, then the arc of the wheel.
