if you have the regular cab configuration, the two vents are to allow the air pressure that is created when you close your door to escape. you would really have to slam the door shut otherwise.
If the vents are inside the cab of the truck, this could mean you have blown your heater core.
Those are VENTS. So when you close the doors with the windows up it lets the air out of the cab. The cab will only hold so much air.

Assuming it is Club Cab, you must first remove the rear seating as well as panels to access them. They are in the rear wall of the truck beneath the panels. I believe this holds true for Crew Cab and Standard Cab.
