If the entire coolant system was drained or parts of it opened it is highly recommended to get the special tool which draws a vacuum then replaces it with coolant. This is because it is easy to airlock many parts of the coolant system.
The amount of coolant it will take to fill your vehicle is dependent upon how low the coolant level is. You can add coolant to your coolant reservoir. The reservoir will have an indicator of when the coolant system is full.

It doesn't. Your rings must be going, or a blow-by. You're lucky the Previa has a back up oil tank unda the hood :)
It isn't going to permanently damage it as long as you replace it with the proper coolant. NAPA makes an all types and models coolant that is compatible with Dex-Cool. Dex-cool just has additional lubricants in it that are designed for that system. If you use regular coolant the seals in the cooling system can break down much faster and cause leaks.
