Jelly is American jam, but in most countries it is a bouncing, sugary desert. Jello is yellow jelly [desert jelly] from the movie Cloudy With A Chance Of Meatballs.

If you mean what you put on a sandwich, that is jelly. The other is often a desert and it is spelled Jell-O.
